There's a six-pointed star in the Great Seal of the United States that's part of the design on the back of all $1 bills issued since 1935, but it's not a Star of David. It's simply one of the ways to symmetrically arrange 13 stars representing the original colonies.

No, the designers of the US dollar bill had nothing to do with Freemasonry. The six-pointed star or double triangle does have meanings in Freemasonry and also is used by Jewish people as the star of David. That is, however, a coincidence. Stars and crescents have meanings in Freemasonry also, and coincidentally also have meanings in Islam. Christian symbols sometimes also have Masonic significance.
The modern US five dollar bill (or note) does not have a red star on it. some older versions did carry a seal printed in red, but this would hardly be seen as a star.
