I guess the study of the introvert or fantasy v reality. Amelie becomes an introvert based on her sheltered upbringing and lack of social interaction. Because she has no friends and after the death of her mother her father becomes even more distant from her she resorts to living inside her own imagination which causes her to romanticise the realities of her life. It's the power of her imagination and her taking joy in the little pleasures of life that most people take for granted which enables her to remain her generally optimistic quirky self as well as instill in herself a desire to help others. However as the film progresses we discover that this desire to become a guardian angel to everyone is actually Amelie's way of keeping herself sheltered and closed off from reality by allowing herself to by living her life and create her fantastical desires through other people. For example it's established early on by the narrator that Amelie is lonely so she creates the relationship between Gorgette and Joseph. Amelie has a hard time expressing herself openly such as calling out Colingnon the grocer about his horrid verbal abuse towards Lucien so she instead resorts to pranks and indirect expressions of her feelings towards him to avoid confrontating him and actually experiencing that interaction herself.

This is why her relationships with the two other main characters in the film- The Glass Man and Nino are so important.

The Glassman is perhaps the personification of what Amelie could become. He is isolated and cannot physically leave his appartment. Maybe Amelie on a subconscious level recognises this which is why he and Amelie become friends they both understand each other. However it is the Glassman's own transformation that triggers Amelie;s. Through Amelie he gets a picture of the outside world that he is denied both through his observations of her and through the video she sends him and in turn the Glassman gives Amelie the encouragment she needs to stop hiding behind her little gestures that allow herself some external satisfaction but leave her feeling as lonely and isolated inside. And Nino is the possibility of what could be. Nino is like Amelie an idealist and a dreamer but he allows himself to both take pleasure in his fantasies and quirks and enjoy the interaction he has with others. Which I think is part of the reason Amelie falls for him. At the end when Amelie finally opens the door to Nino it's her actually opening herself up to someone and allowing them in and thus not only growing confident to pursue an active relationship with someone but also confident in herself and in accepting her need for love and interaction.

So I don't think the film is saying there is a problem with being introverted but it's about being able to find the balance between the two extremes of being really extroverted (take those like Joseph and Colingnon who brashly say what they think with little to no concern for how it will affect others) and too introverted.
