Mainly it depends on how you use it, but most of the time it isn't, if you're just talking about 'the assistant principal.' If it's specifically Assistant Principal Johnson or whoever, then you would.

I heard the man who fell from the roof was once an assistant principal for a high school out west.


Assistant Principal Surname yelled at us and gave us detention.

It usually depends on the situation but whether you capitalize Assistant Principal or not is entirely up to you since it can be capitalized and it can be left not capitalized however you should always capitalize Assistant Principal if you are putting the name of the Assistant Principal either before or after the job title.
