In the 1978 film there is no connection between them, he just takes a bit of a sick shine to her when she drops the keys off at his old house, Halloween II is when her being his little sister is first revealed.

In the original string of films, she is portrayed by Jamie Lee Curtis and she is finally killed by Michael in 'Halloween: Resurrection', and in the remakes she is portrayed by Scout Taylor-Compton, and is shot dead by the police at the end of the second film along with Michael and Loomis, thereby ending Rob's string of films definitively.
