Agar deeps are used to see whether an organism requires oxygen to grow. If there is spreading growth only at the bottom of the tube, the organism is an obligate anaerobe (meaning it cannot tolerate oxygen). If there is growth only at the surface of the agar, the organism is an obligate aerobe (it cannot grow without oxygen). And if there is growth all along the point of innoculation, the organism is a facultative anaerobe and can survive either way.

An agar deep is used to find out the gas requirements of an microbe. Does it thrive in oxygen? Or in oxygen lacking enviroments? After inoculating the microbe in an agar deep, look at the results. If it only grows on the surface, it means the microbe requires oxygen. But if it can also grow within the deep, below the surface, it shows the microbe is capable or surviving in low oxygen concentration. Lastly, if the microbe is found growing far away from the surface, it means that this microbe grows in anaerobic conditions Hope this helps. From, A student studying microbiology
