When Moses held up his hands, the isrealites won. When Moses did not hold up his hands, then the isrealites were defeated. Holding up your hand is a symol of asking God for help. It shows the people that God would give them great victory. Their own strength would not be able to let them win, but believing in God will.

In Deuteronomy 34, we are just told that Moses died and no-one knows where he is buried. The general location is provided as Moab which is near the Arabian desert. There is nothing about an instruction from God.
God could communicate with Moses with a bush in flames that wouldn't go out. He told Moses to help set his people free from the slavery from the Egyptians and also to take his saddles of for that he was standing on holy ground.
