As the Ripper was never identified or caught any motive is pure conjecture
A lot of killers don't understand why they pick the victims they do. Opportunity has something to do with it no doubt. But most go for a certain look. Ted Bundy liked brunettes with long hair. All of his victims were pretty and clean cut looking. They looked like what the they were, college students.
If you look at the pictures available of the Ripper victims, not just the '5' but others that the police knew belonged to the Ripper. Most of them resemble each other. It was a look that appealed to him for whatever reasons. If you look at the pictures of Annie Chapman she resembled many of his other victims. So why he picked her was probably a combination of opportunity and she looked like what he wanted. There were many women in the same sorry state that the Rippers victims were in. Homeless, poor, alcoholic, and for the most part, living on the fringes of society. They made easy prey. And when one turned up dead it seemed no family members were outraged on their behalf. But he didn't take them. He took the ones that fit what he wanted.
Chapman was killed in basically the same way as all the other victims of JTR. The most likely scenario is the killer cut her throat while standing behind her. She had some bruising on her jaw that was probably from the killer clamping a hand over her mouth which would have silenced her and given the killer the advantage of keeping out of direct way of arterial spray. Once she was on the ground the Ripper wasted no time in mutilating the rest of her body.
Since Jack The Ripper was never caught there is no way to know if he even had a wife. If he did, he probably did not kill her or he would have fallen under suspicion immediately. Usually killers like the Ripper will kill strangers not family members.
At the age of 48, she was mutilated and murdered by Jack the Ripper.
In the related links box below, I posted the wikipedia article on Annie Chapman.
Jack struck her acroos the left temple leaving a bruise and he terribly mutilated her body in the throat and abdomen leaving her face swollen after the crime.

The Jack the Ripper deaths occurred in 1888. There is some uncertainty about which murder victims at the time died at the hands of Jack the Ripper, but he is thought to have killed at least three women: Mary Ann Nichols (31st August), Annie Chapman (8 September) and Catherine Eddowes (30 September).
The five canonical victims are Mary Ann Nichols, Annie Chapman, Elizabeth Stride, Catherine Eddows and Mary Kelly.
Consumption, or tuberculosis as it is now called, was rampant in London's impoverished east end. Ripper victim Annie Chapman, upon autopsy, was discovered to have tuberculosis and possibly phlueresy.
