A centrefire cartridge has a primer centred at the rear of the casing. This primer contains a charge which is ignited when the primer is struck (normally by a firing pin, though some guns will have the pin integral with the hammer), and this ignites the gunpowder whidh propels the bullet forward.

Rimfire guns work in pretty much the same fashion, except that they don't use primers. Rather, the priming charge is packed into the rim of the cartridge.
