On both occasions the Paleozoic Era and the Mesozoic Era ended with a mass extinctions.

The Paleozoic ended with the Permian-Triassic (P-Tr) Extinction. This is widely recognised as the worst mass extinction of all time. It is also known as the "Great Dying". Up to 96% of all marine species, 70% of all land vertebrate species and also it is the only known mass extinction of insects with 57% of families and 83% of all genera became extinct. It also finally ended the dominance of the Synapsids (mammal like reptiles).

Similarly the Mesozoic Era ended with a mass extinction, the Cretaceous-Tertiary (K-T) Extinction. Famous for the death of the all non avian dinosaurs, pterosaurs, mosasaurs and plesiasaurs along with many other species of plants and animals.
