The Mesozoic started after the Permian extinction event and ended with the Cretaceous extinction event. The causes of the Permian extinction event are still being studied, and so are the Cretaceous extinction causes, although likely factors include the Chixulub impact event, coupled with large scale volcanism.
Eras are separated by major events that occurred in these eras. At the end of Mesozoic the dinosaurs were extinct and in the paleozoic there was the Permiam Triassic Extinction
