AFAIK, The University of Maryland, University College is the largest in terms of enrollment. They offer classes at most US military bases all over the world, and have a total enrollment of 80,000 or 90,000 - possibly more.

Texas Tech University in Lubbock, TX is second with over 1839 acres on its main campus there.
Ohio State Columbus as a little over 1700 acres.
EDIT: actually Standford and Duke are both 8000+ acres. Texas A&M is bigger than both Tech and OSU, the College Station, TX campus is around 5,200 acres since the whole city is basically the university.
