The difference between premier district and regional is simply area defined. District involves schools and associations in a small area. Regional is defined as a broader area often defined as a large part of the state.
Typically regional is a higher position than district. Traditionally regionals have a wider or more physically expanded area than a district manager. This varies by company though.

Regional manager is the head of a region, say North Region - where he is in-charge of the entire operations and under him there could be many area managers who take care of the area specific operations like in UP-west area, Chandigarh area, etc. The regional manager does not get involve in the small day-to-day issues and manages the entire region and reports to the HQ. The area manager reports to the regional manager and his KRA includes the area specific targets, unlike regional manager who is answerable to HQ if the regional performance is not good.
