Usually, yes. Copying a file is just hat: making another copy of it. Backing it up assumes that you are putting it in a safer place than the original. If you copied a file but kept the copy on the same hard drive as the original, that would not be considered a backup.
It depends on how the term is used, but they both are essentially the same thing. Also, if you are talking about some item that is copyrighted, a DVD, CD, sheet of music or something like that, copying and backing up are almost always illegal. Under most circumstances, you are not allowed to reproduce anything like that for any reason.
