Musical - is a theatrical presentation with songs where the emphasis of the story telling is usually conveyed through song much more than regular dialog. This is not always the rule, but pretty much, and until recently Musicals have tended to be lighter in theme Play (or Straight-Play, which means it has no music, not that it's just for straight people) is a theatrical presentation that doesn't use music to tell the story, etc. There are many shows that confuse the two distinctions and blend them, but at a base level, there you go. Musicals are often much more elaborate and expensive to produce and tend to run longer than straight plays.
Opera has little to no speaking. Most the time, no talking whatsoever, all singing. A play, I assume you mean musical, has much more of a script. The scores' music only interjects throughout the story, instead of carrying it. Musical Theatre requires a major acting background, Opera requires an extensive singing background.
