It is the opposite of despite (even though), so it means "becauase" or "due to".
This word still isn't in microsoft's dictionary, unfortunately, so many people don't use it. Try not to use it too much, because one day you will come across a teacher/boss/mean granny who does'nt think that the word exists.

"Despite of" is not a grammatically correct phrase. Perhaps you mean "In spite of" or simply "Despite." "In spite of" is an alternate for "Despite".
