Pyruvate is used to make acetyl CoA, for instance for the Krebs cycle. Each pyruvic acid molecule is broken down to form carbon dioxide and a 2-carbon acetyl group, which enters the Krebs cycle. Pyruvic acid is converted to acetyl coenzyme A with production of CO2, as welll reduction of nicotinamide adenine dinucleotide to NADH and H+ (and addition of coenzyme A).

Pyruvate dehydrogenase catalyzes this oxidative decarboxylation of pyruvate to form acetyl CoA. The core of the complex consists of 600 copies of E2 (dihydrolipoyl transacetylase, a subunit enzyme whose prosthetic group is dihydrolipoyl transacetylase). At the periphery are 30 copies of £1 (a tetramer), 12 copies of E3 (dihydrolipoyl dehydrogenase, an enzyme whose prosthetic group is flavin adenine dinucleotider) plus 12 copies of an E3 binding protein which links E3 to E2.
