The significance is the following:

1) When lots of stars are plotted in the HR diagram, the vast majority star appear along a curve. This curve is known as the main sequence.

2) It turns out that those are basically the stars that burn hydrogen-1, converting it into helium-4.

An H-R diagram is a graph of stars' luminosity (left side of scale) or absolute magnitude (right side of scale) versus its temperature (at top of scale, hottest left to right) or color (bottom of scale, whiter to the left and red to the right) determined by its relative color and luminosity when compared to the sun of our solar system. In an H-R diagram the sun is given a value of 1 in luminosity and .66 color index (or 5780 Kelvin).
