The exact job is, sadly, unknown. Some time between 1585 when the twins were born and 1592 when Greene lampooned Shakespeare and paraphrased a line from one of Shakespeare's plays, Shakespeare moved to London and became an actor and playwright, working with one or more companies in one or both capacities.
Shakespeare's first job in London in 1586 was said to be a man-of-all-work about theaters. This job was basically to hold horses at the door of the theater for patrons. Later in the theater world this job was changed to the name of 'Shakespeare's boys.'
There is no real record of what William Shakespeare's first occupation was, but it is safe to say that he probably worked in the family trade business. There is little to be known about Shakespeare between the time that he left school and moved to London and when he returned to Stratford-upon-Avon.
