They speak Nihongo (Japanese-日本語) which is their native language. Some English is spoken in tourist areas.

The main language in Tokyo, or Japan in general, is Japanese. Many people know English to some extent, but few are fluent. There are also a handful of Japanese who know Chinese and Korean.

Other languages spoken by Japanese people are quite rare. Foreigners are exceptions; Chinese, Korean, Indian, German, French, Italian, you name it.
Japanese is the only official language of Japan, and the only language widely spoken in all parts of Japan. Additional languages spoken by individuals will vary across the board, depending on what languages they have chosen to study.

Almost everyone knows little to a lot of English because they start learning it in the second year of junior high school (7th grade).

The language most widely spoken in Tokyo is Japanese.

English, Korean, and various dialects of Chinese are also spoken by many, but more often in private than in public. (Chinese, Koreans, and persons from countries dominated by English comprise the majority of non-Japanese living in Tokyo, and many signs and public services allow or have some use of Chinese, Korean, or English).

Tokyo is a very large city, however, so it is possible to hear scores, if not hundreds, of other languages used, including, if you want to stretch the usage of "spoken" to include it, shuwa, or Japanese sign language.
