In the very first Star Wars film, Princess Leia's ship was under attack and the first line was said by C-3PO when he and R2-D2 were trying to get away. An explosion happened and 3PO went: "Did you hear that? They shut down the main reactor! What are we gonna do? We'll be sent to the spice mines of Kessel, smashed into who knows what!"

If you mean the first line of the movie commonly known as "Star Wars" (i.e.: Episode IV), in which case it's CPO (played by Anthony Daniels):

Did you hear that? They've shut down the main reactor, they'll be destroyed for sure. This is madness.

If you're talking about the first line of the whole Star Wars series then it still depends on the order you prefer.

If you order by original release date, then it's still the same as above from the 1977 release of Episode IV. However, in true episodic order, then the first line obviously comes from Episode I, which is Qui Gon Jinn (Liam Neeson) addressing the pilot of the Jedi Shuttle:
