One major one is not being able walk or stand up if they have been in space for an extended period of time. This is because they have gotten used to weightlessness in space and muscles atrophy a bit. It does take an extended period of time for this to happen.

The space travel induces some changes in the body which can be potentially lthal to body if not taken care of it properly. The main factor is the gravity change experienced by the body during the space travel which takes time to recover. The astronauts are kept in special pressurized chambers after they return from space journey to avoid any physical damage.

There are a number of challenges and concerns for living in space.

1. Space is a vacuum. There is no air, no oxygen to breathe. We must take our own oxygen with us, and we must be able to filter out or reprocess the waste gasses like carbon dioxide that would certainly kill the crew if not kept under control.

2. There's no food in space. Astronauts must take their own food and water.

3. The different gravitational pulls and the lack thereof also affects humans. Bone and muscle deterioration happens due to this.

4. Heat management: In space, you can't ignore the problem of either getting rid of or retaining the heat in the spacecraft.
