Pick them up by using three fingers (thumb included) to gently pick them up by the BASE of their tail (if you pick them up by the end of their tail, it hurts them and their tail may break off). Handle them quickly and DON'T get nervous or hover your hand over them for ages before picking them up, as this makes them nervous (speaking from experience here).
Place them on the palm of your hand, and keep GENTLY holding their tail if you're worried you'll drop them.
They usually feel more comfortable when you use both hands to gently lift them from underneath their torso, making sure they feel stable. It is never a good idea to pick them up by their tail or scruff of their neck as it could cause bodily injury and emotional stress.
Well, you should never pick them up by their tails, as this would cause discomfort. The way I pick up my rat is i slide my hand gently under his back feet. (make sure you don't push or pinch their feet in an akward way!) Then, I slide my other handunder his front feet, and make a platform for him/her with your hands. after they are out of the cage, hold them to your chest, sit them on your lap, just make sure their back feet are always supported. NEVER pick a rat up by its underarms, as you would when picking up a baby. This would cause discomfort to the rat and may rip a tendon. Rats are not very fragile, but don't squish them. You do not have to treat it like its made of breakable glass. Just make sure your rat is comfortable.

You must go very slowly and only hold it for a few minutes at a time until it starts to recognize that you're not going to hurt it. Keep your voice down and be very gentle with the rat. Move slowly so you don't startle it.
