people turning into werewolves is just a myth, but if you were to turn into one, your bone structure, skin size, and MANYother things would change so I can imagine it would be very painful!
Although this is not known as a fact, in my opinion yes it does. In films werewolves are always howling when they transform, this could be a sign of pain or discomfort, also the way the fur pierces through the skin looks though it would cause much pain.
On the theoretical side
Transformation from a human state (bipedal, humanoid) to werewolf state (quadrupedal, wolf/human hybrid) would be enormously painful.
In just the head alone the changes are staggering.
The front of the face would elongate into a muzzle.
The dentition would be very different, wolves are carnivores, humans are omnivores not to mention the changes in the soft pallet and tongue (see throat below).
The ears would move to the top of a much narrower head (there's a headache waiting to happen).
The eyes would move out to the sides of the head.
The throat would alter, wolves do not have the complex structures that allow speech).
I could go on, but I think I've made the point that the transformation (if it were anything but fictional) would be painful enough to drive the subject mad (if they didn't die from the structural damage)

Well first you have to actually find both a vampire and a werewolf, then you have to get to know the vampire well enough for them to want to change you, then they'll change you. Then you have to wait until the werewolf you've found turns into a wolf, and then you have to get its attention so it'll cut/scratch/maul you. And there you go - now your a vampire-werewolf. Odd combination. Good luck. Oh, and make sure you don't get murdered brutally in the process, could be quite painful.
