Chronologically, the first time he uses a whip is when he was a young man. In Indiana Jones and the Last Crusade (1989), young Indiana uses a whip to fend off a lion he encounters on the train in the first scene. However, it is in Raiders of the Lost Ark (1981) where he first uses a whip. He uses it in the first scene to grab the revolver out of the hand of one of his guides just before entering the cave.

We see in the original trilogy of films that Indie received his whip and hat after an incident on a train during his youth. The train was being used to transport zoo animals and after falling into a compartment with a lion, he uses the whip (which he finds lying around, miraculously) to fend it off. This is how he got his whip.
