If you have the 7MGE engine: Then the knock sensor is screwed into the engine block, directly underneath the intake manifold.
Knock sensor is attached to the engine side facing the firewall. It's not easy to reach.
Bank 2 Knock Sensor under intake manifold block, near the passenger side. Bank 1 is near driver side
Knock sensor is located in the wall of the engine block facing the firewall. You have lift the car if you want to reach it.

It is on the engine, under the intake manifold, there should be a wire going to a sensor there. It is right in the middle.
It should be in the side of the engine facing the firewall.
I own an 02 Highlander, and sometimes I get a knock when engine is under load, like going up a hill or when accelerating. This is engine knock caused by low octane gas. When I use a mid grade or premium octane, I do not get the sound at all. .
Unless you are skilled in working with engines, I would advise you to bring it to a mechanic. If you are, get either a Hayes or Toyota manuel and go step by step to taking apart your engine. The knock sensor is in the exact middle of the engine.
