Possibly it is when Abraham prayed for an heir Gen 15:2-3.

Genesis 15:2 But Abram said, "Lord GOD, what will You give me, seeing I go childless, and the heir of my house is Eliezer of Damascus?"

Prayer is simply talking to/with God. It is inferred that Adam and then Eve had a continual and relaxed or uninhibited times of prayer. It wasn't until the Fall in Genesis 3:8-9(a) that this 'shameless and friendly' talking ended and the banning from the Garden of Paradise and the need to be humbly request many things began.

The first mention of prayer is in Genesis 4:26. It says that men began to call on the name of the Lord. When we call on the name of the Lord it is prayer.

My trusty copy of Strong's tells me that:

'Pray' appears first (in the KJV) in Gen 12:13.

'Prayer' is first seen in 2Sa 7:27.
