This refers to the time when a young woman was to be stoned (illegally) by a lynch mob because the crown had claimed that she had committed adultery. By making this statement Jesus made them all realise that all of us are sinners. At the end, no one was left to condemn the woman, Jesus sent her on her way with the words 'Neither do I condemn you - Go - but sin no more...' The whole story can be found in John's gospel Chapter 8, with the actual words of the question in John chapter 8, verse 7.
In the specific instance, Jesus spoke to a crowd that had come after a woman caught in adultery to stone her for her sin according to religious law. Jesus' point was, which one of you in this crowd is so good and so pure, that you can legitimately condemn this woman for her sin? Let that one step forward and throw the first stone. Of course, some young hotheads were about to do it, but the older folks held them back, the crowd broke up, and gradually everyone went on about their business, leaving the woman with Jesus. It is then that Jesus tells her, "Is there no one left to condemn you? Neither do I. Go, and do not sin again." That is grace!

Then Jesus replied let those without sin cast the first stone. After all the bible says we have al sinned and fallen from the glory of God.
