Fannie Eilshemius Hesse, the New Jersey -- born wife of Walther Hesse, one of Koch's assistants, had learned of agar from a Dutch acquaintance and suggested its use when she heard of the difficulties with Gelatin. Agar -- solidified medium was an instant success and continues to be essential in all areas of microbiology.

The growth media such as nutrient agar or Mckonkey agar is used to isolate microbial cells from the mix culture when inoculate on them. Some time speical media is used which selectivly grow some time of bacteria or a differential media which give different morphology of different bacterial species e.g. blood agar and Mannitol salt agar. It can be done by three ways: the spread plate method, the streak plate method, and the pour plate method.
By studying, I would hope. Microbiology is a course based solely on the unseen, it really pays to have hands on experience with microscopes, agar plates, and other tests to see it in action. A solid background in biology will help you pass a course on microbiology, but expect to learn a lot more in detail about things like mitosis, cell walls, protein formation, cellular respiration, and more.

Mannitol salt agar or MSA is a commonly used growth medium in microbiology. It contains a high concentration (~7.5%-10%) of salt (NaCl), making it selective for Staphylococci (and Micrococcaceae) since this level of NaCl is inhibitory to most other bacteria.
