Macbeth didn't think Macduff was a threat because the witches said "None of woman born shall harm Macbeth." Macbeth welcomes this good news and, assuming Macduff was born the natural way, Macbeth thinks he has nothing to fear.

Macduff had fled to England but he left his wife and kids behind. Macbeth had been told by the witches to "beware Macduff" so he he wanted macduff out of the way. Macbeth sent his soldiers to kill Macduff but when they arrived at Macduff's castle to find he had fled they killed his family instead. This meant the soldiers could go back to Macbeth with a result and Macduff didn't have an heir.

Macbeth orders the death of Macduff because he is told by the witches to "beware Macduff" when he goes to see them. Macbeth sends the two murders who he sent to assassinate Banquo and Fleance to murder Macduff, but when they get there, Macduff has already fled to England to get Malcolms help. The murderers kill Lady Macduff and Macduffs son because Macbeth plans to seize Macduffs castle and because he no longer has much of a conscience and can not distinguish between right and wrong anymore.

Not for a good reason. He finds that while he has been consulting the witches, Macduff has fled. Being frustrated and angry (since Macduff is the fellow he is supposed to beware), he wants to attack him indirectly. It is an irrational and emotional act.
