By his 'diction' I think one means Twain's writing style. He had a wonderful ear for dialect (for example, look at the way Jim speaks) and his deceptively simple use of language makes the novel accessible - that is, direct and easily understood.

He was deliberately turning away from European literature and rebelling against what he considered was the convoluted style of novelists like Sir Walter Scott - in fact, Twain calls the wreck of the boat where murder is being planned 'Sir Walter Scott'.

Writing in the first person - seeing the world through Huck's eyes is part of Twain's style or diction too, and it gives a vividness and simplicity - Huck is just a boy with no great education - to the novel.
