If the work is done properly the values for the controls will be the same or within an expected range of variance as the known concentrations. The values of the real samples are then likely to be correct. This is a quality control or quality assurance procedure. Alternately the same samples can be sent out to a number of labs (Real and control samples). The results should be consistent. This procedure would identify labs that have consistently higher results for all samples. This process is called Round Robin testing
