This is an interesting question to get in a job interview - it basically asks, what is your biggest weakness that will be a problem for the company?
If you have any criminal convictions, this would be an excellent time to mention them - owning up to time in jail for a DUI, minor drug charge or a stupid teenage prank that went wrong will show that you are honest. Once you admit to the conviction, however, you should go on to emphasize that this was a previous mistake but you have taken steps to ensure it doesn't happen again (got sober, went through rehab, did your time but have been law-abiding since, etc.).
Other things you may want to note: if you've had attendance issues, if your skills are weaker than expected, if you will need additional training. All of these could be problems for the hiring company, particularly if you try to hide them. Be honest about your background and say, I don't have a lot of experience working with Microsoft Office, but I am willing to learn.
