He has been declared a bastard son and therefore Don Pedro becomes the King rather than Don John, and this gives Don John deep cause for resentment. It is not his fault he is considered an unlawful son and yet he has been discriminated against all his life for it. Also, in the start of the play he has just lost his rebellion against his brother and although Don Pedro forgives him this only makes Don John angrier as he is forced to live under his brother's thumb. "If I had my mouth, I would bite". In short, Don John is sick and bitter from being treated and labelled a bastard and missing out on his rights, he also shows some jealousy of Claudio and he resents not being able to be his "evil" self. He acknowledges that he is evil and he likes the fact, and his long festering bitterness at his treatment propels him to try and ruin the happiness of others.
