Prince Charles is next in line to the throne of Great Britain and Northern Ireland, if he survives the Queen. However, there is talk of the monarchy being passed over one generation in William's favor. A special act of Parliament would need to be passed if this were to happen.
If Charles dies before William, yes.
Otherwise, if Charles says he does not want it. It is unprecidented and I personally think, Charles would want to give William time to spend with his wife and perhaps children as much as he could.

... it goes on from there to Anne's children and grandchild, and then to Charles' cousin, son of Elizabeth's sister Margaret, and then his descendants, but it's unlikely that that many people will die in quick succession.
When William and Kate have a child, that child will become third in line (assuming no one higher on the list has died yet) and push Harry and everyone below him down one. If it's a girl, then she'll still be third in line, but she'll be pushed down to fourth if they later have a son (all sons come before all daughters, which is why Anne, who was born before Andrew, is tenth on the current list instead of fourth).

If Charles dies before Queen Elizabeth dies, then Prince William will be first in line. If Charles dies while he is king (having succeeded Elizabeth to the throne), then Wlliam becomes king and his eldest son (supposing there is one) will be heir to the throne. If William does not have children, then his younger brother Harry will be first in line.
