I believe that they did not. In fact, they even had follow movies where Matt is alone and was never with Miss Kitty.
James Arness , Amanda Blake , Milburn Stone and Ken Curtis .
Matt caught her in Docs office in bed with Doc and Matt said GET OUTTA DODGE.
All kidding aside people, Amanda Blake and Kilburn Stone really did have a long term term relationship going on in real life.

As far as I know Kitty Wells never was an actress in Gunsmoke. There was a Miss Kitty played be Amanda Blake. Kitty Wells was a Country Music singer in the 50's, 60's and 70's.

I just watched season 4 "The Coward" and Matt and Chester were looking for a man in the different saloons in Dodge and the ones they showed were called, "Texas Trail", "Trivoli", "Lady Gay", "Longbranch", "El Dorado", and "Buffalo Lager". She did not quit. She was fired by Executive Producer John Mantley. They had ongoing disputes over wardrobe and she disrespected him in front of the cast and crew while filiming the episode, The Lost. When her contract came up for renewal, he let her go.
The late Milburn Stone (July 5, 1904 - June 12, 1980) who played in the role of "Doc Galen Adams" made his last appearance in the "Gunsmoke" series in the episode "The Sharecroppers" (31 March 1975) . I remember that he was absent from the series due to having heart surgery but I couldn't find which episodes in which he was absent then returned .
Actor Pat Hingle , playing in the role of "Dr. John Chapman" , made his appearance in 1971 ( Gunsmoke: Season 17, Episode 11 - "Drago" [22 Nov. 1971] ) which was about the same time the late Milburn Stone was temporarily replaced so I'm guessing it was in 1971 .

The Gunsmoke television series was filmed on a studio backlot in Hollywood, CA. While the series occasionally shot on location, most of the outdoor sets were located on what is now called CBS Studio Center at 4024 Radford Avenue, in the Studio City area of the San Fernando Valley. Previous to CBS taking it over, the lot had been used by the Republic Pictures studio, which is best known for many action adventure serials and westerns. Earlier, it had been the location of the Mack Sennett studios, famous for the Keystone Kops silent film comedies. The exterior scenes for the Gilligan's Island series, around the lagoon, were also filmed on this studio lot, but the lagoon was paved over and is now the site of a parking garage.
