No you cannot. Luckily, you are given a warning before you attempt the Hoover Dam quest. Save your game in a separate slot before going to Hoover Dam and you should be able to continue it after you've beat the game. There may be DLC that allows you to continue the game after the final quest, but it's yet to be released.
