For the most part, girls were not allowed to be knights. I can think of one exception, Joan of Arc, a young french girl who fought wars, was burned at the stake, and became a saint. Kights were from noble familys.

Another example can be seen in the women of Tortosa, a town in Catalonia. There was a battle there, in which the women of the town, acting on their own, attacked and routed an enemy army. They were all made knights. They were freed from all taxation, and given the right to lead processions, to speak before the men in public assemblies, and to be treated in all other respects in a manner befitting of their rank.
In the Middle Ages, you generally had to be born a lady; becoming a lady was rare.
