It's impossible to say, without knowing exactly where in Brooklyn you're coming from. Brooklyn is almost 100 square miles in area and almost all of New York City's subway lines run through it. The directions will be completely different, depending on where in Brooklyn you're coming from.

The only subway lines that stop at 42nd Street-Grand Central Station are the 4-5-6 (the dark green line) and the 7train (the purple line). Of these, the 4 and 5 are the only lines that run into Brooklyn. Unless you are starting out near one of the stops on the 4 and 5, you will have to transfer.

It depends where you are going to, as Brooklyn is a large borough. You can go to mta.info and go to trip planner and find out where you are going. It will give you trip info and any delays.

Take the Downtown 4-5-6 (the dark green line) from 42nd Street-Grand Central Station to Brooklyn Bridge-City Hall. You will want to take the 4 or 5, though, not the 6, as the 4 and 5 are express trains and it's only 2 stops from 42nd Street-Grand Central to Brooklyn Bridge-City Hall.
