Wall studs are to be 16" on center in most places.

20 feet x 12 inches (# of inches per foot) = 240 / 16 (spacing of wall studs) = 15

15 x 2 (both of the 20 foot walls) = 30

30 x 12 = 360 / 16 = 23 x 2 = 46

Add 4 extra for the corners so 4 + 46 + 30 = 80.

That's just the walls. The rest will depend on any doors or windows you have because those will require adding extra jack & king studs to carry the load as well as how you decide to handle the roof.

One hundred eighty linear feet makes your perimeter. Doubling up for a sole plate doubles that. The top plate adds that number again. Using 12- 14- or 16-foot long 2x4s changes the equation. The allowances for windows and doors also complicates your issue. If your room will have 2x4 underpinnings below the floor, and ceiling rafters, there are more 2x4s to add. Good luck!

Builders will normally order 1 per foot when framing 16" on center to cover the cripples and corners. This does not include the wood required for the bottom and top plate.

figure 1 per foot so 30 + 30 foot of stud for bottom plate = 60 foot of stud for top plate .... if using 8ft lumber i would get 42 total.
