The answer to that question is that Jacob has always been a Werewolf. He was supposed to be the pack leader.He was born a werewolf but have not yet found the trigger. According to the Quileute legends he turns into a werewolf simply because the "vampires" were near.It was in his blood and already apart of him other wise he wouldn't be the ere or Emphrian or that he was supposed to be the pack leader because of his heritage. The reason they decided to find the trigger was because of the vampires

In the series, the people who are in the "pack" often refer to themselves as "Wolves" instead of werewolves, and it is stated clearly that they are shapeshifters who theoritcally could take the form of a different animal if they possessed the abilities of their ancestor Spirit Warriors.
Because the Indian ancestry of the Quileutes that Jacob is apart of are known to be in relation with wolves,so it runs in the blood and that's how Jacob is a werewolf because its in his blood he was born that way.If i am correct he is the only one in the wolf clan that is a werewolf by blood.
