First and foremost, Columbus, because he discovered the New World and that is what led to the colonization of New Spain.

Secondly was Hernando De Cortez, who conquered the Aztecs and therefor brought the land that would become New Spain under Spanish rule.

Charles V of Spain was the one who began the colony, and Antonio de Mendoza became its first viceroy (somewhat like a governor).
