'Playthrough 2.5' is not a new game mode, it is how the game is referred to once you complete your second playthrough. It changes the game so that the enemy levels are scaled to yours.

No. You have Playthrough 1, and then Playthrough 2. Once Playthrough 2 is completed, the game is in what is called Playthrough 2.5 which is not a new playthrough, it just increases enemy levels further if you want to replay a previous area.
You can not change your character in mid game, if you want to play a different character you need to start a new playthrough, the game you abandon will be saved so you can return to play it if you choose too.
You can start a Playthrough 2 where you will complete each mission again, but all the enemies are at a higher level, starting from around Lv33. Otherwise there isn't much to do except buy some of the DLC packs.
After you beat the game, you have a choice to play either Playthrough 1 which is where you continue on after you beat the game to beat other uncomplete missions, or Playthrough 2 which is where you start from the beginning on your same character after the bus takes off and you meet the Claptrap. On Playthrough 2 the enemies are a higher level (should be similar to you characters level) and you get better weapons, you get to keep your level, weapons, shields, etc. Sadly, there is no Playthrough 3 (I've beaten the game on Playthrough 1 and 2 on my Siren, plus all the Downloadable Content, so now I'm at max level 69). Yes you can, you need to have played the game far enough to power up the Fast Travel system, and of course, you need DLC pack 1 purchased and installed. If all's done correctly, you can travel to Jakob's Cove to start the DLC content.
Playthrough 2 is a feature in Borderlands that allows players who have completed the game's story-line to start the game over again with the same character on a more advanced mode.
