Although this was placed in the marijuana category along with others, I assume by "painkillers" you are referring to medical opiods (such as codeine, hydrocodone). The answer is yes; if you are tested during or soon after opiod use, you will likely fail. However, the general procedure for drug testing usually includes "Are you taking any medications?" etc, at which point you would say "Yes, I've been prescribed (prescription here)" optionally followed by "for (ailment)". If you aren't asked this, you will likely be able to explain your failure as being caused by your prescription.
