What are Ambrisentan tablets?

AMBRISENTAN is used to treat a serious heart and lung disorder called primary pulmonary hypertension. This medicine may help to increase your ability to exercise and improve your breathing.

This medicine may be used for other purposes; ask your health care provider or pharmacist if you have questions.

What should I tell my health care provider before I take this medicine?

They need to know if you have any of these conditions:
• liver disease
• an unusual or allergic reaction to ambrisentan, other medicines, foods, dyes, or preservatives
• pregnant or trying to get pregnant
• breast-feeding

How should this medicine be used?

Take this medicine by mouth with a glass of water. Follow the directions on the prescription label. You can take this medicine with or without food. Take your doses at regular intervals. Do not cut, crush or chew this medicine. Do not take more often than directed. Do not stop taking except on the advice of your doctor.

Talk to your pediatrician regarding the use of this medicine in children. Special care may be needed.

A special MedGuide will be given to you by the pharmacist with each prescription and refill. Be sure to read this information carefully each time.

What should I watch for while taking this medicine?

Visit your doctor or health care professional for checks on your progress. Despite receiving this medicine, your condition may worsen and you may need your dose adjusted. Call your doctor or health care professional if your conditions gets worse.

Pregnancy must be avoided while taking this medicine due to the potential for serious side effects to an unborn child. If you are female of child-bearing potential, you will have to have pregnancy tests regularly (at least monthly) while you are taking this medicine. Discuss options to prevent pregnancy with your doctor or health care professional.

Breast-feeding should be avoided while taking this medicine. It is not known if ambrisentan can pass through your milk and harm your baby.

• fever
• general ill feeling or flu-like symptoms
• light-colored stools
• loss of appetite, nausea
• right upper belly pain
• swelling of the ankles or legs
• unusually tired or weak
• yellowing of eyes, skin

Side effects that usually do not require medical attention (report to your doctor or health care professional if they continue or are bothersome):
• constipation
• flushing of the skin
• headache
• runny nose or sore throat
• stomach upset

Where can I keep my medicine?

Keep out of the reach of children.

Store this medicine in its original packaging at room temperature between 15 and 30 degrees C (59 and 86 degrees F). Throw away any unused medicine after the expiration date.

Ambrisentan (U.S. trade name Letairis; E.U. trade name Volibris) is a drug indicated for use in the treatment of pulmonary hypertension.

It functions as an endothelin receptor antagonist, and is selective for the type A endothelin receptor (ETA).[1]

Ambrisentan was approved for sale by the U.S. Food and Drug Administration (FDA) on June 15, 2007 for the once-daily treatment of pulmonary arterial hypertension.[2][3][4] It was later approved by the European Medicines Agency for use in the EU on April 2008.[5] Ambrisentan had previously been designated an orphan drug by both the FDA and the European Commission, in August 2004 and May 2005 respectively.[6]

Clinical uses

Ambrisentan is indicated for the treatment of pulmonary arterial hypertension (WHO Group 1) in patients with WHO class II or III symptoms to improve exercise capacity and delay clinical worsening.

The LETAIRIS Education and Access Program

The LETAIRIS Education and Access Program (LEAP) is a program to help physicians and patients learn about the risks of LETAIRIS, including the serious risks of liver injury and birth defects.

LEAP works by:

  • Providing information to prescribers on the risks of LETAIRIS
  • Providing comprehensive education to patients and assistance with obtaining LETAIRIS
  • Requiring enrollment of both prescriber and patient in LEAP
  • Controlling dispensing through a specialized distribution network (specialty pharmacies)
