abbr. for paired box; one of a family of transcriptional regulators that contain a paired-box domain and are specifically expressed during the development of a wide range of structures and organs. There are nine Pax genes in mouse and in human. They are expressed primarily in the neural tube and brain in the early embryo. Pax1 and Pax9 are closed related, as are the groups Pax3 and Pax7, and Pax2, Pax5, and Pax8. PAX2 protein (392 amino acids) is also expressed in the eye and kidney; mutations cause the renal-coloboma syndrome. PAX3 (836 amino acids) controls MyoD expression in limb buds; numerous mutations cause Waardenburg syndrome. PAX6 (422 amino acids) is identical in sequence in mouse and in human, and contains a homeobox domain and a C-terminal region rich in proline, serine, and threonine. It is also expressed in eye and pancreatic islet cells. Numerous mutations cause aniridia. PAX8 is implicated in embryonic development of the thyroid gland; mutations cause thyroid dysgenesis and congenital hypothyroidism.
