150 grain Remmington Core-Lokt is a good round.
SHOT PLACEMENT IS THE MOST IMPORTANT FACTOR; any load of at least 130 gr will get the job done.

General rule of thumb is that the best caliber is the one which best suits you. Of the two, .270 would be the more powerful round. Both are substantial for hunting something like deer, although the .243 is probably better suited for the experienced hunter, whereas the .270 would be a little more forgiving to the novice hunter. The .270 is a necked down .30-06, and the .243 is a necked down .308.

Generally speaking, no. You can only fire the caliber for which the gun is chambered. There are some exceptions, but not many.
They are two different (but related) calibers of cartridges. The .270 is basically the 30-06 cartridge that has been necked down from .30 to .27 caliber. The "rate of twist" for the rifling will also be different, since you are shooting a skinnier, less heavy bullet.
