There is no such thing as a 'best animation' course, because everyone requires certain elements from their tutor - that is of course, meaning that people learn differently. The best way to learn is to (whether it is traditional animation, stop motion or computer animation) practice it. If traditional, draw with construction. If it is with a computer, use appropriate software. And if with stop motion, experiment with the models that you have.

All animation would have characters whether it be a person, a tree, or a cup of tea (no, seriously). Having a well designed character is important as you will be working with that character for many hours of the week while practicing your animation (as you may want to work on it through a short). Though, I recommend practicing animating with a well designed, well known character as those characters will be ones that have stood the test of time.

Since you are not specific in the type of animation you want to learn I can only say that animation is a broad subject. You could use a mouse, a pencil, your hands, paper cut outs, and the list goes on and on. Be specific in your next question and you may get it answered in a less ambiguous manner.
