The flight time is not different, and if it is, that is because of the direction the plane is flying. There is a jet stream (strong wind) that blows from the East (Johannesburg) to the West (Perth), so it would seem to make the plane go faster.
The shortest routes are to fly Perth to Johannesburg or Sydney to Johannesburg. There is no route by road.
It has to be quicker to fly from the UK because Florida is on the opposite side of the planet from Perth.

No. I've not heard of any flights that go directly between Washington DC and Johannesburg but I imagine the flight will fly east over the Atlantic, then so a sharp curve southeast towards the Canary Islands and West Africa, then I think the flight will turn south over Nigeria and then head directly south to Johannesburg.
