The answer is definitely "yes." Your brain actually develops a tolerance to Adderall by creating more receptors. Once you develop a tolerance, the Adderall becomes less and less effective. You can overcome the tolerance temporarily by increasing the dose but, in time, your brain will also develop a tolerance to the new dose.

Amphetamine is a central nervous system stimulant which tolerance is built to extremely quickly, especially when the user dose escalates on his/her own, and does not follow directions from their doctor. The more amphetamine you take, the more tolerance you will build, and the less it will be effective. Amphetamine also has cross-tolerance, a phenomena not seen in most other drugs; side effects such as paranoia increase with extremely high dose use.
