Yes we can. The primary responsibility is to the safety of the passengers and secondary is to the equipment we fly. If safety is an issue we are allowed to vary from the flight plan. This information is relayed to dispatch and to ATC. The normal reason for variance from the planned flight is for health reasons of passengers. The second most common reason is for weather. When the issue is weather related, schedules become an issue as well as the ability to safely store equipment on the ground. Weather, for example, can cause us to land at airports that are not the intended airport or the point of origination. Normally only part 91 flying varies with any regularity. Professional pilots tends to fly craft that has more capabilities then the general aviation portion of the flying public.

It all depends upon what type of flying the pilot is doing. In general all pilots can change their route at anytime for any reason.

An Airline Transport Pilot flying for the airlines will generally only change their route for the reasons stated above, i.e. someone gets sick or injured and they need to land to get them help. Other reasons might include changing route to avoid bad weather.

A Commercial pilot, Private pilot, Recreational pilot or sport pilot can and will change there route at any time for whatever reason they see fit. As a Private pilot myself, i regularly change my route just because I changed my mind as to where I wanted to land or the weather forced me to change where I am going.
